DKkaraoke has a very irritating lead-flute for inexperienced singers to follow, I have been trying in vain for weeks on and off to remove it with different sound editing programs. Does anyone know any good trick for this?

There isn't much you can do, it's encoded in the disc as part of the music - which is why many do not like the DK series. You could try & eq it out, but you will be taking out the music as well at those frequencies.

Lonnie expressed my sentiments to the letter.
I find the lead flute on DK to be extreamly irritating, but most inexperienced karaokers don't really notice. It's the experienced singers that find that it cramps thier style, and most of them cope.
I've tried to EQ it out and it's my opinion that it isn't worth the bother or the diminished audio.
I suggest that you just live with it and keep your singer's favorites in mind when you buy new disks.
